After slumping to 2-4 and 3-43 early, Lanning and Haynes combined for Australia's biggest partnership in a decade with the pair adding 156 since joining at the wicket.
The pair took the game to England in the second session, with Australia able to retain the Ashes for a fourth straight series with a win at Manuka Oval.
The innings has also left both on the brink of their maiden Test centuries, with captain Lanning on 84 and her deputy Haynes on 82.
The pair have both played brilliantly through the offside, cutting when given width and Haynes supplying the shot of the day with a boundary through the covers.
Their partnership is already the 12th biggest of all time in Tests for Australia, and the largest since Haynes combined with Jodie Fields to put on 229 for the sixth wicket in 2009.
England have however had four crucial chances go begging.
Lanning was put down on zero at second slip, and while it was a low chance it would have left Australia in real trouble at 4-43.
She was dropped again on 14 and appeared to edge one behind on 37 but there was no appeal made.
Australia's captain also survived a shout for both caught and lbw on 52, but the third umpire ruled the ball hit her forearm and not her glove and would have gone over the stumps.
Hayne's best chance came when she was on 43 and cut Nat Sciver to Katherine Brunt at second slip, but it also went to ground.
The partnership came after Alyssa Healy went for a duck and Beth Mooney was out for three, with the pair both caught behind as Anya Shrubsole and Katherine Brunt had the ball swinging.
Ellyse Perry fell for 18 after cutting nicely in her innings before taking on a short ball from Sciver and being caught behind to make it 3-43.
It marked the first time in six innings and six years that Perry has been dismissed for less than 100 in a Test match.
